New Study Indicates Multiple Mechanisms Involved In The Inhibition Of The Growth Of LNCaP Prostate Cancer Cells By Plant Lignans

Secaucus, NJ – May 2, 2006 – Linnea has announced that new research performed by the Department of Clinical Medicine at the University of Insubria in Italy has shed further light into the multiple mechanisms involved in the inhibition of the growth of LNCaP prostate cancer cells by plant lignans.

Several lines of evidence suggest that plant-derived dietary lignans may play a role in chemoprevention and prostate health promotion, through complex pharmacological activities that may include pro or anti-estrogenic, pro-apoptotic, antioxidant, and anti-angiogenic mechanisms. In particular, in-vivo research had shown the novel lignan, hydroxymatairesinol (HMRlignan), and its human lignan metabolite enterolactone, to inhibit tumor growth rate and tumor take rates in animal models (Bylund et al, Exp Biol Med, March 2005).

In this new study, designed to investigate the mechanisms involved in the modulation of cell viability of the human androgen-dependent prostate cancer, LNCaP cells lines were incubated with either enterolactone (70-100 microMols) or hydroxymatairesinol (70-100 microMols) for 48 hours, with estradiol (100 microMols) and the cytotoxic agent cycloheximide (10 microMols) used as the experimental controls. Results showed a concentration-dependent increase in the proportion of apoptotic cells for both substances -- enterolactone was about two-fold as effective as hydroxymatairesinol in this experiment but less than half that of cycloheximide and estradiol. In these experiments the estrogen receptor antagonist tamoxifene significantly reduced the effects of both enterolactone and hydroxymatairesinol, as well as that of estradiol.
Subsequent RT-PCR assays showed that treatement of LNCaP cells with either enterolactone or hydroxymatairesinol (0,01-1 microMols) also increased mRNA levels for the proapoptotic protein Bax and decreased mRNA levels for thè antiapoptotic protein Bcl-2, thus resulting in an overall decrease of thè ratio Bcl-2/Bax, which can be taken as an index of cell viability. In agreement with this observation, both substances in the same concentration range also increased the expression of the mRNA for caspase-3, which is the common final effector of the intracellular pro-apoptotic cascade.

Commenting on the results, research pharmacologist, Dr. Marco Cosentino, stated, “these results indicate that, in-vitro, the viability of human prostate cancer cells is impaired by the lignans enterolactone and, to a lesser extent, hydroxymatairesinol. The effect is, however, milder than that of an established cytotoxic agent such as cycloheximide, and is likely to be exerted at least in part via estrogen-like mechanisms. This is the first study showing the ability of enterolactone and hydroxymatairesinol to modify the intracellular balance between pro- and anti-apoptotic proteins, an action which likely contributes to the overall pro-apoptotic effect of both lignans. Such an action is, however, exerted at concentrations that are several tenfold lower than those affecting cell viability, suggesting that additional mechanisms are involved in the final pro-apoptotic effect. On the other side, one could also speculate that the ability of enterolactone and hydroxymatairesinol to affect the intracellular mechanisms regulating cell death and survival may also result in ’sensitization’ of the cells to the effects of other antiproliferative agents.”

Cosentino added, “Our data support the notion that hydroxmatairesinol, and to a greater extent its human metabolite enterolactone, may suppress the growth of prostate cancer cells. Also in view of its well-established tolerability and bioavailability, hydroxmatairesinol represents a viable dietary supplement providing a suitable source for endogenous entrolactone that, in turn, may play a role in the promotion of prostate health.”

LOCARNO, SWITZERLAND & EASTON, PA - April 28, 2006 - New research performed by the clinical medicine faculty at the University of Insubria in Italy has added to the body of evidence supporting the use of lignan phytoestrogens to help women manage menopause. In the study, designed to establish the effective median concentration value (EC50) of Norway spruce-derived lignan, hydroxymatairesinol and its human metabolite enterolactone in comparison to estradiol, experiments confirmed a daily dosage 10 mg - 30 mg as sufficient to raise circulating enterolactone to estrogenically active levels.

In the study, in which all three compounds displayed estrogenic activity in the MCF-7 breast cell model, estradiol displayed the highest potency in the pM concentration range whilst enterolactone exerted activity in the same concentration range as estradiol but with far less efficacy (about 2/5 that of estradiol).Hydroxymatairesinol exhibited a significantly lower efficacy (about 7/10 that of estradiol) but only in the microM concentration range.

The study concluded that both hydroxymatairesinol and its metabolite enterolactone are endowed with estrogenic activity, which is likely to be exerted through estrogen receptors and to target the same intracellular mechanisms acted upon by estradiol. The estrogenicity of hydroxymatairesinol and enterolactone is however milder than that of estradiol, as indicated by the lower potencies and efficacies of both lignans.

Commenting on the significance of the study the lead research pharmacologist, Dr. Marco Cosentino, stated, 'The present results support the notion that dietary supplementation with Norway spruce lignans provides a suitable source for endogenous enterolactone which in turn is likely beneficial for several conditions related to estrogen insufficiency (e.g., bone, andcardiovascular disturbances, as well as menopause-associated symptoms), and at the same time devoid of the adverse effects induced by stronger estrogenic agents. Bioavailability studies in humans show that, after oral supplementation with Norway spruce lignans, plasma concentrations of hydroxymatairesinol and enterolactone occur in the sub-microM range, which are below the observed in vitro EC50 ( effective median concentration) of hydroxymatairesinol but well above the EC50 of enterolactone. Therefore, in humans, hydroxymatairesinol at the currently recommended doses represent mainly a source of enterolactone, which in turn exerts a mild estrogenic activity.

Lignans and Breast Health
There has been extensive research demonstrating the correlation of enterolactone in women and the risk of breast cancer. In a case-control study to examine the association between serum enterolactone and risk of breast cancer in Finnish women researchers analyzed 194 breast cancer cases (68 pre-menopausal and 126 postmenopausal). The study found an inverse association between serum enterolactone and risk of breast cancer among both groups.1 Proprietary research into hydroxymatairesinol (HMRlignan™) and enterolactone supports this finding. In a study conducted at the University of Turku, enterolactone was shown to inhibit the growth of 7,12-dimethylbenz(a)anthracene-induced mammary carcinomas in rats.2 In another study HMRlignan™ was demonstrated to both inhibit tumor growth and to reduce tumor take rates.3

Lignans and Menopause
Estrogen acts by binding to its receptors, and enterolactone may offer a natural selective estrogen-receptor modulator effect. By binding the estrogen receptor, enterolactone may exert an agonist or antagonist response. Depending on the concentration of endogenous estradiol and the distribution of estrogen receptors, enterolactone may act as either an estrogen receptor agonist or as an estrogen antagonist. When estrogen levels fall enterolactone may exert a weak estrogen-like effect (mimicking the presence of estrogen). Conversely, when estrogen levels are high, enterolactone will occupy and thus block estrogen receptors, thereby moderating the significant hormonal fluctuations that give rise to the typical symptoms of menopause. Healthy levels of circulating enterolactone may decrease both severity and occurrences of uncomfortable symptoms women experience.

“The good news for women is that the estrogenic activity of enterolactone is relatively weak and therefore the risk of unwanted effects due to adverse estrogenicity is minimal,” says Ward.

Lignans and Heart Health
Research into diet and associated risk factors of cardiovascular disease has also been a focus of investigations in both men and women. The Kuopio Ischaemic Heart Disease Risk Factor Study, a population-based study of middle-aged Finnish men on the associations between serum enterolactone level and the risk of coronary heart disease (CHD) and cardiovascular disease (CVD), concluded that high serum enterolactone level is dose-dependently positively associated with reduced CHD and CVD-related mortality4.

Again specific research into hydroxymatairesinol (HMRlignan™) and enterolactone lends support to these findings. In a Finnish study to evaluate evidence that low serum enterolactone concentration might be an independent risk factor for acute coronary events, researchers proposed that due to the bi-phenolic structure of enterolactone, along with the parent lignans, could act as an antioxidant and thus contribute to cardiovascular health. The study involving 100 participants tested the hypothesis that a low serum enterolactone concentration is associated with increased in vivo lipid peroxidation. In a multivariate analytical model, enterolactone persisted as a significant predictor after adjustment for vitamins and other variables, with the strongest associations with F2-isoprostanes. The study results support the hypothesis that low serum enterolactone concentration is associated with enhanced in vivo lipid peroxidation.5 Proprietary research focusing on hydroxymatairesinol and enterolactone provides further pharmacological evidence on the benefits of HMRlignan™ for heart health. In vitro tests show HMRlignan™ to be a strong antioxidant more or equipotent to Trolox, to incorporate into isolated LDL fractions, and to protect from copper-induced LDL oxidation in vitro.3 Ongoing research also indicates the capacity of HMRlignan™ and enterolactone to modulate inflammatory reactions in heart health.

Introducing HMRlignan™: A Novel Enterolactone Precursor
and More Digestible Source of Lignans

On April 19, 2005, the USDA unveiled a new Food Pyramid. In the USDA’s introductory report, entitled, “Dietary Guidelines for Americans 2005,” under chapter 5, “Food Groups to Encourage,” the USDA places emphasis on consumption of whole grains. It reads, “In the grain-refining process, most of the bran and some of the germ is removed, resulting in the loss of dietary fiber (also known as cereal fiber), vitamins, minerals, lignans, phytoestrogens [emphasis added by author] phenolic compounds, and phytic acid.”

This report recognizes the value of the broad range of phytonutrients now missing from the average American diet due to overprocessing of foods or poor diet selection. While most Americans recognize the need for a healthy diet and the consumption of fibers, lignans and other phytonutrients, changes to dietary habits are not always easily achieved. For many people the concept of a daily bowl of flax seed is less than appetizing. However, HMRlignan™, a new generation low-dose, highly bioavailable lignan supplement, may provide a more digestible alternative.

An increasing pool of scientific knowledge and strong epidemiological data has raised the profile of lignan-rich sources such as flax with their relevance in health maintenance being recognized by consumers and dietary supplement formulators alike. Due to the extensive educational and marketing campaign efforts by the flax industry the most recognized lignans by the public are those found in flax seeds. However, lignans are an abundant phytonutrient found in a wide variety of foods and plants, notably unrefined grain products, seeds (sesame and flax), berries, fruits and vegetables. Upon ingestion, lignans make their way into the gastrointestinal tract whereby the microflora convert them into “human lignans,” primarily enterolactone, which exerts the weak estrogen-like activity that is primary in their reported prophylactic action in protecting against cardiovascular disease and tumors.

Although lignans are abundant in unprocessed foods, obtaining a sufficient daily dose remains a challenge. Flax seed is used often to supplement the daily intake, but quality and standardization are problematic, relative bioavailability is low and stability is an issue particularly in lignan rich unrefined oils that are prone to rancidity and subsequently carry a short shelf life. In such an environment, the lignan quality can be greatly compromised. The side effects from high consumption of flax seed can also be uncomfortable, including a pronounced laxative effect, bloating and flatulence. In addition, the high daily dosage, at 20 to 30 grams daily, presents a major challenge to supplement formulators.

Although these challenges exist for flax seed, 7-hydroxymatairesinol (HMRlignan™) is the first commercially available lignan source to overcome these hurdles. 7-hydroxymatairesinol is the prevalent lignan in sesame seeds, but is also particularly abundant in the Norway spruce (Picea abies). The Norway spruce is plentiful in the Nordic countries such as Finland, where it is used extensively in the paper industry (a primary local industry), which has led to a strong national industrial and academic interest in the study of wood chemistry.

In addition, Finland is also a center of excellence for the study of phytoestrogens and it was researchers at the University of Helsinki who made the link between the research chemistry and the potential human health benefits of the wood derived lignan 7-hydroxymatairesinol (HMRlignan™) and its metabolism to the protective weak phyto-hormone and lower incidences of breast and prostate cancers and cardiovascular disease. This led to the partnership between Hormos Medical Corporation of Turku, Finland, and Linnea, Switzerland, and the success in bringing HMRlignan™ from the academic arena to the consumer.

In 2003, the Kuopio Ischaemic Heart Disease Risk Factor Study looked at the associations between serum enterolactone levels and the risk of coronary heart disease (CHD) and cardiovascular disease (CVD) in middle-aged Finnish men. The conclusion was that high serum enterolactone level is dose-dependently positively associated with reduced CHD- and CVD-related mortality in this population.

A 2002 cross-sectional study (Framingham Offspring Study) in the U.S. investigated whether dietary estrogens have similar beneficial effects on metabolic cardiovascular risk factors as estrogen therapies in 939 postmenopausal women. In the highest quartile of isoflavone intake, the mean cardiovascular risk factor metabolic score was 0.43 lower than the lowest quartile. The difference in this score between the extreme quartiles of intake of lignans was 0.55 points. The study concluded that high intake of lignans in postmenopausal women was associated with a favorable metabolic cardiovascular risk profile.

Studies also support an inverse association between elevated enterolactone levels and rates of breast and prostate cancer. Breast cancer is considered to be a hormone-regulated disease with the female sex hormone estrogen known to increase the risk of development, through a complicated series of pathways.

Enterolactone binds weakly to the estrogen receptors, and as a weak estrogen, appears to block overt estrogen activity in specific selected tissues. In addition, enterolactone stimulates the synthesis and circulating levels of a biochemical called sex hormone-binding globulin (SHBG). Through this activity, enterolactone appears to reduce the free bioavailable pool of circulating estrogen, thereby reducing estrogen penetration in tissues and diminishing risk for adverse estrogen balance. Third, there is evidence that enterolactone may also inhibit biosynthesis of estrogen by blocking aromatase, a key enzyme in biosynthesis of estradiol. Collectively through multiple mechanisms of action, lignans appear to positively influence optimal estrogen balance in the body.

In prostate health, an increasing body of evidence points to the benefits of lignans in the maintenance of prostatic wellness, and suggests that enterolactone may have direct inhibitory effects associated with cancer cell growth and signaling. Research has shown that enterolactone may compete with E2 for the type II estrogen receptor, induce sex hormone binding globulin and may also play a role in steroid metabolism and synthesis, thus reducing proliferation of hormone-dependant prostate. A 2005 study investigated the effect of 7-HMR on LNCaP human prostate cancer xenografts in mice. Results showed that a 7-HMR diet significantly inhibited the growth of LNCaP tumors. Mice that ingested 7-HMR had smaller tumor volume, lower tumor take rate, increased proportion of non-growing tumors and higher tumor cell apoptotic index compared with controls.

Lignans, like 7-hydroxymatairesinol (HMRlignanTM), can also make a contribution to the management of menopause. Enterolactone, by binding the estrogen receptor in different target tissues, may bring about either agonist or antagonist type response, depending on the availability and recruitment of tissue-specific co-regulatory proteins to the estrogen receptor signaling complex. When estrogen levels decline, enterolactone may exert a weak estrogen-like effect, mimicking the presence of estrogen). When estrogen levels are high, enterolactone occupies and blocks estrogen receptors, thereby acting to smooth the peaks and valleys. During the climacteric (the several-year period when estrogen levels are in flux, leading to cessation of menstruation) estrogen levels are in flux, so a formula that addresses the body’s own production of enterolactone may indeed help lessen symptom severity and frequency.

Bone health is also an emerging area where lignans are being shown to play a positive role. Here too, estrogens are known to play an important role in bone maintenance. Recent studies have shown a link between lignan consumption and bone reabsorption.
